There is a handy chart circulating on Reddit that shows the unlock path to every character. Say you really wanted to unlock Lil Mac from Mike Tyson's Punch Out. You'd begin your trek toward unlocking Lil Mac by playing with Mario. Mario unlocks Sonic, who then unlocks Bayonetta, who in turn unlocks Lil Mac. You would essentially follow this pattern to unlock every character in the game.
